The separation of complex lipide mixtures by the use of silicic acid chromatography.

چکیده
It is now well established that the various lipide classes of blood (1)) liver (2)) and other organs are metabolically dissimilar. The different fatty acids of the diet (3) and within the organism (4) also have been shown to have variable functions and effects. Methods have recently become available for the rapid and precise analysis of these fatty acids (5) and it is reasonable to expect that an analysis of the fat,ty acid composition of each lipide class will yield rich information concerning the transport and metabolism of fatty acids. Considerations of this type indicate the need for a reliable, standardized method for the separation of the lipide classes. The present communication describes a method for such separation based on adsorption chromatography with silicic acid. Since Trappe’s introduction of silicic acid chromatography for lipide separation in 1940 (6, 7), a considerable experience has been gained with this procedure (8-11). Such information has been put to practical use in many types of lipide separations (1, 12, 13) but workers have found that the separations can be capricious and unreliable. Therefore, a systematic study of the many variables seemed desirable. Information obtained from this study and from previous investigations of the properties of silicic acid (14-16) were used to elaborate a standardized method for the separation of lipide groups. The present report is a description of the method and some representative results.

Observations on the silicic acid chromatography of the neutral lipides of rat liver, beef liver, and yeast.
Within the past several years the application of silicic acid to the chromatographic fractionation of simple and complex lipides has received increased attention. In particular this adsorbent has been used in the separation of phospholipides by McKibbin (I), Lea et aE. (Z), and Hanahan et al. (3). As shown by Borgstrom (4), who used model lipide compounds, and Fillerup and Mead (5), who employe...
متن کاملThe separation of sterol intermediates in cholesterol biosynthesis by high pressure liquid chromatography.
A three-step procedure has been developed for the separation of complex mixtures of sterol intermediates in cholesterol biosynthesis. The method has been applied to the separation of sterol intermediates formed from [14C]mevalonate by normal rat hepatocyte culture cells. In Step 1, a short gravity-flow silicic acid column (1.2 x 6.5 cm) separates the incubation products into four classes consis...

bUnequivocal class separations of tissue lipids are important particularly whcn high temperature gas Chromatography is used at the same time for the determination of the long-chain acid composition of major lipid classes. This problem was studied most recently by Hirsch and Ahrens (1) ; their procedurc involves silicic acid column chromatography with clution by ether-petroleum ether mixtures fo...

s i l i c i c acid column chromatography has proved very useful in the separation and isolation of many of the sterols found in the nonsaponifiable fraction of lipid mixtures (1-8). In the course of an investigation of the nonsaponifiable lipids formed from mevalonic acid-C14 by a rat ascites tumor, a highly radioactive fraction was isolated by chromatography 011 silicic acid. This fraction was...
متن کاملSeparation of Glycosyl Diglycerides from Phosphatides Using Silicic Acid Column Chromatography.
A procedure has been developed for the separation of the glycosyl diglycerides from the phosphatides of Gram-positive bacteria on columns of silicic acid. The method utilizes mixtures of acetone in chloroform for elution of the glycosyl diglycerides, followed by increasing amounts of methanol in chloroform for elution of the phosphatides. The course of the fractionation was followed by means of...
